Mass spectrometry is a powerful analytical technique that can accurately determine the mass of compounds and perform qualitative and quantitative analysis based on the unique mass fingerprints of different compounds. It has extremely important applications in many fields such as chemistry, biology, medicine, and environmental science.

Its working principle is based on ionizing sample molecules, and then using the motion characteristics of ions in electric and magnetic fields to separate and detect ions according to the mass-to-charge ratio (m/z). The specific process is as follows: First, the sample is introduced into the ion source through the injection system, and the ion source converts the sample molecules into gaseous ions. There are many types of ion sources, such as electron impact ionization source (EI), electrospray ionization source (ESI), matrix-assisted laser desorption ionization source (MALDI), etc. The EI source bombards the sample molecules with a high-energy electron beam, causing them to lose electrons to form ions; the ESI source converts the sample molecules in the solution into charged droplets under the action of a high electric field, and eventually forms gaseous ions as the solvent evaporates; the MALDI source uses a laser to irradiate a mixture of the sample and the matrix to ionize the sample molecules. Next, the ions are accelerated and enter the mass analyzer, which separates them in space or time according to their different mass-to-charge ratios. Common mass analyzers include quadrupole mass analyzers, time-of-flight mass analyzers (TOF), and ion trap mass analyzers. The quadrupole mass analyzer applies specific DC and RF voltages to enable ions of a specific mass-to-charge ratio to pass through the quadrupole stably, thereby achieving ion separation; the TOF mass analyzer separates ions based on the different flight times of ions in the field-free flight space, and the flight time is related to the mass-to-charge ratio of the ions; the ion trap mass analyzer captures ions in the trap through an electric field, and then changes the electric field parameters to allow ions of a specific mass-to-charge ratio to be ejected from the trap and detected. Finally, the separated ions are detected by the detector, which converts the ion signals into electrical signals, and after amplification and data processing, a mass spectrum is obtained. The mass spectrum uses the mass-to-charge ratio as the horizontal axis and the ion intensity as the vertical axis. The ion peaks of different mass-to-charge ratios represent different compounds or compound fragments. By analyzing the mass spectrum, the chemical composition and structure of the sample can be determined.

The mass spectrometer is mainly composed of an injection system, an ion source, a mass analyzer, a detector, and a data processing system. The injection system is responsible for introducing the sample into the ion source. Depending on the state and properties of the sample, the injection system has various forms, such as direct injection, gas chromatography injection, liquid chromatography injection, etc. As mentioned above, the ion source is a key component for ionizing sample molecules. The mass analyzer and detector complete the separation and detection of ions respectively. The data processing system processes, analyzes and stores the signals collected by the detector, analyzes the mass spectrum through special software, identifies compounds, and performs quantitative calculations.
Depending on the mass analyzer, mass spectrometry is mainly divided into quadrupole mass spectrometry, time-of-flight mass spectrometry, ion trap mass spectrometry, Fourier transform ion cyclotron resonance mass spectrometry (FT-ICR MS), and other types. Quadrupole mass spectrometry has a simple structure, low cost, fast scanning speed, and is widely used in routine analysis; time-of-flight mass spectrometry has the characteristics of high resolution and high mass range, and is suitable for the analysis of macromolecular compounds; ion trap mass spectrometry can perform multi-stage tandem analysis on ions to obtain more compound structure information, and is often used in proteomics research; FT-ICR MS has extremely high resolution and accuracy, and can accurately analyze complex mixtures, but the instrument cost is high and maintenance is complex.

Mass spectrometry has a wide range of applications in many fields. In the biomedical field, it is used in proteomics research. By performing mass spectrometry analysis on protein enzymatic products, the types and sequences of proteins are identified, and protein modifications and interactions are studied, providing important basis for disease diagnosis and drug development; in drug development, it is used for the analysis of drug metabolites to understand the metabolic pathways and processes of drugs in the body. In the field of environmental science, it is used to detect pollutants in the environment, such as volatile organic compounds in the atmosphere, pesticide residues in water, heavy metals in soil, etc. Mass spectrometry analysis can accurately determine the types and concentrations of pollutants and evaluate environmental quality. In the field of food safety, it is used to detect additives, pesticide and veterinary drug residues, illegal additives, etc. in food to ensure food safety. In the field of materials science, it is used to analyze the composition and structure of materials and study the relationship between the performance and structure of materials.
